Named after landowner Jeptha Hughes, Hughesville had grown up around a gristmill constructed in 1820. Hughesville became a town to help the local farmers and citizens of eastern Lycoming County. Some of the industries that came to the town included a chair factory, a wagon shop, several distilleries, and a furniture factory. Each year, Hughesville plays host to the Lycoming County Fair.
